Here are some screenshots I took while I was making this. I apologize for the inconsistency or if you feel like they do not clearly reflect my work process. I tend to get into a flow and forget to keep track of taking screenshots of the progress.  I have written down how I went with this artwork but if you any questions feel free to contact me (addresses given below).
I began by dividing the original map by a grid and started drawing the artwork based on an 'isometric grid'. This helped maintain the ratio and estimate the sizes and distance between objects when drawing from a flat 2D grid to an isometric 3D grid. I resized the isometric grid when I drew smaller objects like the buildings and homes. Even smaller when I drew the bicycle, trees, and bushes. 

Later, I realized some changes had to be made now that it is in 3D and seen from a different angle (30º). Wherever there was a jumping cliff, that meant an elevated surface. These elevations were not visible in the original map because we are looking straight head, from down to up, in those map. Imagine you are in a moving car and in a distance is a tall building with four sides. The moment your view aligns straightly with the building, you only see one side of it i.e. the side facing you (this is what happens in the original map). And as you keep going, then you begin to see the other side as well. In our case, the elevation becomes visible.

Moving ahead, I had finished the map but it looked bland with plain solid colors. So, to ensure the isometric artwork resembled the atmosphere of the original map, I developed custom pattern swatches and pattern brushes based on the element's from the original map. I made use of these to give texture to the trees, mountain, river, green grass, etc.

And lastly, since these are vector based, they can be resized to any size. Here are two close-ups:
